Overview
The Measurements tool provides interactive line and region-of-interest measurements directly on the current image display.
X pixel and Y pixel specify the physical size represented by one image pixel in each direction.
Available units are px, nm, µm, mm, cm, and m.
Lock X=Y keeps both pixel dimensions identical and is appropriate for square pixels.
Apply calibration stores the selected calibration for subsequent measurements.
Clear calibration returns measurements to pixel units.
Draw a line ROI to measure length and inspect intensity along the selected path.
The line profile opens in a separate window and updates when the line ROI is moved.
With calibration active, distance is shown in the selected physical unit; otherwise it is shown in pixels.
Rectangle, Ellipse, Polygon, and Freehand tools calculate statistics within the selected region.
Reported values include mean, standard deviation, minimum, maximum, pixel area, percentage of image area, and calibrated area when calibration is active.
Right-click an area ROI to open additional commands: Histogram, Update Statistics, and Delete ROI.
The histogram displays the intensity distribution of pixels inside the selected ROI.
Use the Bins slider to change histogram resolution.
Clear Measurements removes all measurement ROIs, their labels, and associated line-profile windows.
This does not modify the underlying hyperspectral dataset.
Measurements are calculated from the image currently shown in the main Image Display.
If the displayed image is RGB, it is converted to grayscale before intensity statistics are calculated.
Interaction With Real-time Spectra
Starting a measurement disables the active real-time spectrum interaction to avoid competing mouse callbacks during ROI drawing.
1. Display the image or band you want to measure.
2. Enter spatial calibration if physical units are required.
3. Choose the desired ROI type.
4. Draw the ROI on the main image.
5. Move or resize it as needed and inspect the updated measurements.
6. Right-click an area ROI for histogram or other commands.
7. Use Clear Measurements when finished.
Measurement accuracy depends on the validity of the spatial calibration and on the image currently being displayed.
After spatial binning, resizing, geometric transformation, or other resampling, verify that the calibration is still appropriate.
Intensity measurements describe displayed image values and should not automatically be interpreted as concentration or other physical quantities without suitable calibration.
